Is there potential for herbal medicines in amyloid treatment?

Protein function is dependent on the 3-dimensional folding, and proteins failing to adopt the predetermined structure are remover by proteasomal degradation. Failure to remove misfolded proteins can result in the accumulation of proteins, causing disease development. One large group of protein misfolding diseases is amyloidosis, where misfolded proteins form highly ordered fibrillary structures. Today, more than 35 different proteins have been identified as amyloid-forming in humans, and each protein is related to a specific disease. Amyloid diseases can be divided into systemic and localized forms. In the former, amyloid deposits occur throughout the body, while in the localized form are the deposits limited to the site of synthesis. Protein AA is N-terminal fragments of Serum Amyloid A (SAA). AA amyloid is an example of systemic amyloidosis developing secondary to chronic inflammatory diseases, e.g., TBC, rheumatoid arthritis. In contrast, examples of localized amyloid proteins and associated diseases are protein A? that deposits as amyloid in the brain of patients with Alzheimer's disease, Islet amyloid polypeptide (IAPP) forming amyloid in the islets of Langerhans in patients with Type 2 diabetes. The formation of amyloid is cytotoxic to the cells, and depositions result in organ failure.
Professor Yanli Liu is a pharmacologist at the Dept. of Pharmacognosy, Soochow University. In collaboration, we aim to investigate if anemoside B4 and derivatives thereof inhibit amyloid aggregation.
Amyloid-forming IAPP, and AA will be used for in vitro and in vivo analyze the inhibitory effects on protein misfolding. Techniques such as recombinant protein expression, western blot, Tht Assay for kinetic studies of amyloid formation, cell culture, FACS analysis of ROS production, transmission electron microscopy, and confocal microscopy will be used for the project implementation.
