Immunomodulatory effects of GABA on human T lymphocytes

The ?-aminobutyric acid (GABA) is the main inhibitory neurotransmitter in the nervous system playing an important role in the control of neuronal excitability. Interestingly, GABA also has immunomodulatory effect in the immune system and specific GABA receptor are expressed in various immune cell types. The modulatory effect of the GABA system in immune cells is relevant to autoimmune diseases including Type 1 Diabetes (T1D). Repurposing clinically approved GABAergic drugs may provide great opportunities to treat these diseases. The aim of this project is to investigate the plasticity of GABA signaling in human T lymphocytes under physiological and pathophysiological conditions. The regulatory rules about lab safety and working with human blood samples will be instructed. Peripheral blood mononuclear cells (PBMCs) and specific subtypes of T cells will be isolated from fresh blood samples or buffy coats of healthy donors and T1D patients when needed. A variety of experimental methods including proliferation assay, quantitative PCR, Western blot, flow cytometry, immunocytochemistry, patch-clamp recording and Ca2+ imaging will be performed when appropriate. This project will further deepen our understanding of GABA-related signaling pathway networks in immune cells.
