On the influence of powder amount during compression for the prediction of tabletability of binary powder mixtures

Background
One important tablet quality attribute is the tablet tensile strength. This is often product specific and depends on the composition of the powder blend and the pressure applied during manufacturing. The relationship between tablet tensile strength and compaction pressure is commonly denoted a tabletability relationship. By using a three-stage model, tabletability of single component powders can be predicted with a hybrid approach based on some compression parameters generated from one single compression experiment. As pharmaceutical products generally are composed of two or more components it is thus of interest to study the applicability of the approach for binary mixtures. Moreover, as the amount of powder during formulation development often is limited it is of interest to investigate if predictions using compression parameters assessed from a small amount of powder can be used.

Aim
First, the aim is to study the possibility to predict tablet tensile strength for binary powder mixtures using a three-stage compaction model describing the strength-pressure relationship. Secondly, the aim is to investigate the accuracy of performing predictions based on powder compression data assessed using a small amount of powder. Thirdly, the aim is to investigate the possibility to predict tabletability relationships for mixtures based on compression parameters for the single component powders.

Expected outcome
It is expected that the study will bring knowledge on the applicability of the hybrid prediction approach on powder mixtures. Furthermore, the study will give information on the precision of the prediction performed using compression parameters generated from a small amount of powder.
