Physiologically based pharmacokinetic modelling of antibiotics used in sepsis

Sepsis is a life-threatening condition that occurs when a dysregulated host immune response to infection develops. The profound pathophysiologic alterations occurring in sepsis result in high variability in the pharmacokinetics of administered antibiotics with wide variations in drug exposures following fixed dosing. Patients with low drug exposures are less likely to have a positive clinical outcome. However, the understanding of the underlying factors that contribute to this variability is still poor.

This project is aiming to implement physiologically based pharmacokinetic modelling (PBPK) to describe the pharmacokinetic changes of antibiotics in septic patients compared to healthy individuals. This could be used to predict the tissue concentration at the target site and guide clinicians in finding the proper dosing regimens in septic patients.

The project will focus on one of the commonly used antibiotics in sepsis and literature data will be extracted for this purpose. The candidate for this project is expected to have a good understanding of the concepts of pharmacokinetics. Knowing how to use R software is preferable.

he work will be carried out in Uppsala, however in a research group, that offers an international environment. The working language, including supervision, will be English.
