During HT 2024 develop standard powder compositions with lactose and without lactose with suitable powder flow properties which, after the addition of API, can theoretically be filled according to the intermittent dosator capsule filling method.

Background
Recurring problems when filling capsules with the capsule filling machine IMA Adapta 50, where the powder mixture did not have sufficient flow properties or required a lot of compression to be able to form stable powder plugs during capsule filling. These problems have created a need to develop capsule compositions more easily with powder flow properties that have been identified as ideal for this capsule filling machine and that cohesiveness can be controlled.

My task
• Literature studies
- Study the subject (powder flow properties, capsule filling etc.)
- Map raw materials that are commonly used (traditionally used compared to modern ones), their properties etc.
- Is it possible to predict flow characteristics of the composition after addition of API?
- Map the compositions of APL's existing capsule products
o Properties of excipients
o API structure

• Map response to powder plug test versus need for compaction for APLs intermittent dosator capsule filled products (laboratory work)
• Develop standard composition based on APL's excipients which after addition of API meet identified ideal flow properties for capsule filling with IMA Adapta 50 (laboratory work)
- Trials according to design of experiments (DoE) to optimize the properties of the composition
• Develop standard composition based on new excipients that which after addition of API meet identified ideal flow properties for capsule filling with IMA Adapta 50 (laboratory work)
- Trials according to design of experiments (DoE) to optimize the properties of the composition
• Methods of analysis – Different standard powder analysis
