Exploration of different Miltefosine dosing regimens for visceral Leishmaniasis in children for novel dosage forms and schedules

Miltefosine is the only oral drug approved for the treatment of Leishmaniasis in its different forms, the focus of the project will be on Visceral Leishmaniasis. This is a life-threatening diseases that affects mostly tropical areas of the world and is often neglected. Since its introduction the biggest issue has been achieving appropiate exposure in children compared to adults. Recently an allometric dosing reliant on three variables (height, sex and weight) and even more recently a weight band-based have been proposed. The available dosage forms in the market are a 10mg and 50mg capsules that must be taken daily, however they have different complications when implemented in the areas where they are most needed.

The goal of this project is to explore the posibility of a new solid dosage form that can be given to children (from 1 y/o) that are also convenient in both administration frequency and schedule. This work is a simulation model that can propose novel forms while keeping the drug exposure and safety at acceptable levels. The project relies solely on simulations, as the results are expected to awaken interest the future development of an actual dosage form. The end result would include the development of a shiny app. The project will mostly be worked on R, but NONMEM knowledge is valuable.
The work will be carried out in Uppsala, however in a research group, that offers an international environment. The working language, including supervision, will be English.
