Aim
Evaluate which model diagnostics are currently used in the field of PBPK modelling.
Background
During the last 5 years, almost 1500 articles on physiologically based pharmacokinetic (PBPK) modelling have been published. This is a staggering number of publications in a field that keeps growing each year. Sugano et al (2021) published an informative article on the requirements of scientific writing of PBPK modelling. The FDA and EMA have guidances on what should be included in PBPK models and their reports. Interestingly, none of these documents describe the evaluation of PBPK model outcomes in detail. For pharmacometrics, there are some guidelines on how to evaluate the models. For example, the papers by Karlson et al. (2007) and Keizer et al. (2013). We are now wondering which diagnostics researchers in PBPK modelling are using and how suitable these are for the described purpose.
Methods
First, you will be reviewing published PBPK model papers to create your dataset of used diagnostics in PBPK modeling. You will extract data on model diagnostics, evaluations and statistics. Once the dataset is complete, you will evaluate which diagnostic methods are commonly used and what this diagnostic describes. Do these diagnostics describe the objectives, such as goodness-of-fit. Are there better diagnostic methods available that should be used instead?
